I. Overview

An online biosensor is constructed for fermentation processes to obtain the most critical, direct, and efficient real-time biochemical information using key sensing technologies.
A biosensor is a biochemical analysis technology based on biomolecular recognition, using bioactive materials as sensing elements combined with physical or chemical transducers.
Its working principle: enzymes catalyze the conversion of substrates into products; the products are detected by a transducer and converted into quantifiable signals, allowing the concentration of the target substrate to be measured.
Through long-term R&D, the team has developed a new series of fully automatic online biosensors, enabling unattended automatic sampling, measurement, and feedback feeding control of key biological parameters during fermentation.

II. Key Features

The SBA-66 Online Fermentation Analysis System is an online biosensing analyzer using immobilized enzymes as core components.
It integrates automatic sampling, automatic dilution, and automatic measurement for fermentation processes, with one channel of feedback control.
The system supports three biosensing channels.
  • Sampling and measurement cycle: < 10 minutes

  • Repeatability near standard concentration: error < 3%

It can automatically measure glucose, lactate, glutamic acid, ethanol, lysine, and other parameters during fermentation, and control substrate glucose concentration via feedback regulation.

    Sampling-to-measurement cycle < 2 minutes; minimum sampling interval: 10 minutes.
  2. Automatic online dilution:
    Automatically dilutes based on analyte concentration, supporting online measurement of substrates/products up to 300 g/L (glucose, lactate, glutamic acid, glycerol, ethanol, etc.).
  3. High-stability fused enzyme membrane:
    Newly developed by the team; service life > 3 months under continuous sampling; total test capacity > 3000 samples.
  4. High accuracy and precision:
    Error < 2%.
  5. Sterile sampling option:
    Ceramic membrane sterile sampling available, eliminating contamination risk.
  6. Feedback feeding control:
    Automatically adjusts feeding based on substrate/product concentrations for fully unattended operation.

IV. Application Cases

4.1 Laboratory & Pilot-Scale Process Development

The SBA-66 online fermentation system supports process development and scale-up from lab to pilot scale, especially feedback-controlled feeding at fixed glucose concentrations.

4.2 Online Analysis for Enzyme Reagent Fermentation (Unattended Operation)

The system enables online glucose analysis during enzyme reagent fermentation.
Real-time glucose monitoring supports feedback glucose feeding, significantly improving fermentation efficiency and enabling low-carbon-source fermentation.
A glucose feeding control strategy was implemented based on real-time measurements:
glucose concentration maintained at 0.1–0.5 g/L, ensuring sufficient carbon source for cell metabolism while keeping feeding within an optimal range.

4.3 Online Analysis & Automatic Feedback Feeding for Amino Acid Fermentation

Online glucose analysis was applied in amino acid fermentation, with glucose controlled at 4–6 g/L.
Sampling interval: 10–30 minutes.
The system adjusts feedback feeding and links with pH control and ammonia addition, achieving precise glucose control during amino acid synthesis.
